The free amino acid, with no protecting groups on the amine, the carboxyl or the side-chain hydroxyl. It is the starting material from which the protected serine building blocks used in peptide synthesis are made: Nα-acylation with Fmoc-OSu or Fmoc-Cl under Schotten-Baumann conditions gives Fmoc-Ser-OH, and the side-chain ethers and esters are built from there.
All three functional groups are free, so a coupling in which serine is the acyl donor needs the amine and, usually, the hydroxyl masked first; otherwise polymerization and O-acylation compete. The amino acid is zwitterionic and dissolves in water rather than in DMF or dichloromethane, which is the practical reason the protected derivatives, not the free amino acid, are used on the resin.
