H-Gly-2-Cl-trityl Resin
Glycine esterified to 2-chlorotrityl resin, free amine (H-Gly-2-ClTrt); 100–200 mesh, 0.3–0.8 mmol/g
2-Chlorotrityl resin preloaded with glycine and supplied with the amine free, so the second amino acid is coupled directly, without an Fmoc removal. The bulky trityl linker makes diketopiperazine formation at the dipeptide stage less likely than on Wang resin. The loading for each lot is on its certificate of analysis.
The trityl ester is very acid-labile. Dilute acid, such as 1% TFA in DCM or a mixture of acetic acid, trifluoroethanol and DCM, releases the peptide acid with its tert-butyl and Boc side-chain protection in place; 95% TFA with scavengers cleaves the peptide and removes its TFA-labile protecting groups. Because glycine is achiral, the protected fragments can be activated at the C-terminus for fragment condensation without epimerization.
